
Treeline Biosciences has completed a $200 million Series A extension, bringing its total funding to $1.1 billion. The clinical-stage biopharma company announced this milestone alongside the launch of Phase 1 trials for three programs targeting cancer patients.
The funding round attracted significant investor interest, with participation from AI Life Sciences (an affiliate of Access Industries), ARCH Venture Partners, and OrbiMed as lead investors. Notable participants include GV, KKR, accounts advised by T. Rowe Price Associates Inc., Ajax Health/Zeus, Casdin Capital, Fidelity Management & Research Company, Aisling Capital, Rock Springs Capital, and Exor.
The oncology space continues to attract substantial investment as companies target previously difficult-to-treat cancers. Treeline's approach focuses on validated targets that have historically proven challenging to address with traditional drug development methods.

The new capital will support ongoing Phase 1 trials for three programs currently enrolling patients. Two programs target lymphoma patients, while a third addresses solid tumors with specific KRAS mutations. The company plans to use the funding to advance these trials through early clinical milestones.
Treeline also intends to expand its internal R&D capabilities, particularly in computational drug discovery tools that help prioritize development workflows. The funding will support hiring across biology, chemistry, protein science, and clinical development teams operating in the U.S. and Europe.
Josh Bilenker and Jeff Engelman co-founded Treeline, bringing substantial oncology R&D experience to the company. Bilenker previously founded Loxo Oncology, which successfully developed three FDA-approved medicines before its acquisition. His track record in oncology drug development helped attract early investors and talent to Treeline.
Engelman served as director of thoracic oncology at Massachusetts General Hospital before becoming global head of oncology at the Novartis Institutes for BioMedical Research. His clinical and research background complements Bilenker's entrepreneurial experience in building successful biotechnology companies.
The company's approach combines traditional drug discovery methods with advanced computational tools. This strategy aims to accelerate development timelines while maintaining rigorous scientific standards across multiple therapeutic areas including cancer, neurological, and autoimmune diseases.
Treeline has attracted backing from prominent life sciences investors known for supporting companies through clinical development and commercialization. The investor syndicate includes both specialized healthcare funds and generalist investors with significant biotechnology portfolios.
The substantial funding round reflects investor confidence in Treeline's scientific approach and management team. With $1.1 billion raised to date, the company has resources to advance multiple programs through clinical development while continuing to expand its discovery engine and pipeline.
